Abstract
成功将米曲霉(Aspergillus oryzae)乳糖酶基因(lac gene)克隆到毕赤酵母(Pichia pastoris)表达载体中,实现该乳糖酶的有效分泌和高效表达.以米曲霉cDNA文库为模板,通过PCR扩增得到米曲霉乳糖酶基因.将含有该米曲霉乳糖酶基因的表达载体pGAPZαA-lac以电转化方式转入到毕赤酵母表达系统,获得高效表达米曲霉乳糖酶的重组菌株GalC131.该工程菌株在YPD培养基中发酵72 h,发酵液中目的蛋白含量高,乳糖水解酶活达到530 U/mL,并且此工程菌株分泌表达的目的蛋白易于纯化,有利于工业化应用.
