Exploration on the nature of spleen govering transportation and transformation based on the dynamic expression of oatp2a1 in spleen-deficiency rats

Objective:To explore the extensive concept of spleen governing transportation and transformation on the relationship between the expression of oatp2a1 in lung,liver,stomach and small intestine and the metabolism of AA-Ⅰ in spleendeficiency rats.Methods:SD rats were randomly divided into 4 groups:blank group,spleen-deficiency group,blank+AA-Ⅰ group and spleen-deficiency+AA-Ⅰ group.The pharmacokinetics of AA-Ⅰ were made and tissues of lung,stomach,liver,small intestine were taken in blank group and spleen-deficiency group;4 tissues were taken after being given AA-Ⅰ in 5min and 60min in blank+AA-Ⅰ group and spleen-deficiency+AA-Ⅰ group.Expression level of the 4 kinds of tissues in each group was detected.Results:The metabolism of AA-Ⅰ in blank group and spleen-deficiency group was different.The oatp2a1 mRNA expressed higher in lung,liver and stomach tissues in spleen-deficiency group compared to blank group(P0.01,P0.05).The oatp2a1 mRNA expressed lower in small intestine tissue in blank+AA-Ⅰ 60min group compared to blank group(P0.01).The oatp2a1 mRNA expressed lower in lung and liver tissues in spleen-deficiency+AA-Ⅰ 5min group compared to spleen-deficiency group(P0.05).Conclusion:The change of the expression of oatp2a1 is correlated with the metabolism of AA-Ⅰ in spleen-deficiency rats which suggests that lung,liver,stomach and small intestine might play a great role in the function of spleen governing transportation and transformation.
