Abstract
目的:探讨DNA结合蛋白定位技术的改进.方法:以左旋咪唑/酒精先消除细胞的内源性碱性磷酸酶(AP),再分别以DNase和RNase消除细胞内源性核酸,应用低盐浓度的杂交液预杂交后再加特异DNA的标记探针,以链霉亲和素SA-AP中介, 最终以四氮唑盐/5溴-4-氯-3-吲哚磷酸(NBT/BCIP)作为底物, 37 ℃下显色.结果:DBP阳性信号呈紫色细颗粒,主要定位于胞核.结论:此DNA结合蛋白的改进技术定位清晰且染色过程迅速.
