•A calcium phosphate-coated liposomes containing fluorophores was developed.•Distribution of fluorophores in the aqueous core and interface was established by photophysical studies.•Coated liposomes possess a good staining capacity of Staphylococcus aureus biofilm and smears.•The calcium phosphate coating is of critical importance in fluorophore delivery.•The coated vesicles have promising applications in drug delivery.
