•Mouse and rat spermatogenesis were modelled to aid germ cell mutagenicity testing.•The 28 + 28d design provides better spermatogonia exposure than the 28 + 3d design.•Modelling supports the 28 + 28d design as the preferred approach for germ cell testing.
