•The usefulness of the PIGRET assay as a short-term test was assessed.•The genotoxicity of DEN was evaluated using both the RBC Pig-a and PIGRET assays.•No increase in MF was observed in either the RBC Pig-a or PIGRET assay.•The usefulness of the PIGRET assay was suggested by the results of ENU.
