Abstract
目的 研究绿原酸(CGA)对肝星状细胞增殖、胶原蛋白(Collagen)Ⅰ、CollagenⅢ、基质金属蛋白酶抑制剂-1(TIMP-1)及基质金属蛋白酶-2(MMP-2)表达及分泌的影响.方法 体外培养大鼠肝星状细胞株HSC-T6,随机分为5组:阴性对照组、血小板衍生生长因子(PDGF)组、PDGF+ CGA (12.5 μg/mL)组、PDGF+ CGA(25 μg/mL)组和PDGF+CGA (50 μg/mL)组.干预24 h后,MTT法检测HSC-T6的增殖情况;RT PCR检测Collagen Ⅰ、CollagenⅢ、TIMP1及MMP-2 mRNA表达;ELISA检测细胞上清液中Collagen Ⅰ、CollagenⅢ、TIMP-1及MMP-2的蛋白含量.结果 CGA可明显抑制PDGF诱导的肝星状细胞增殖(P<0.05),且随着剂量的增加,抑制作用逐渐增强(P<0.05);CGA可明显抑制PDGF诱导的Collagen Ⅰ、CollagenⅢ、TIMP-1的表达和分泌(P<0.05),但对MMP-2的表达和分泌无明显影响.结论 CGA可通过抑制肝星状细胞增殖及细胞外基质合成、促进细胞外基质降解来发挥抗肝纤维化作用.
