•The chaperoning of parasite derived proteins to the erythrocyte membrane is poorly understood.•HHsp70 interacts with the PEXEL domain containing Type II PfHsp40s but not with the Type I PfHsp40s.•PfHsp70-1 interacts with the Type I PfHsp40s but not with the PEXEL domain containing Type II PfHsp40s.•Potential chaperone-cochaperone complex of hHsp70-PfHsp40 (Type II) is likely to export parasite proteins to the RBC membrane.
