•LPS induces monomethylation of Eno-1 R50 (Eno-1R50me) and thus drives Eno-1 trafficking to the cell surface.•PRMT5 is a novel interacting partner of Eno-1 responsible for Eno-1R50me.•PRMT5 inhibition prevents Eno-1 exteriorization and diminishes the invasive potential of cancer cells.
