•SGE-516 is a neuroactive steroid GABAA receptor positive allosteric modulator.•SGE-516 augments α1β2γ2 and α4β3δ subunit-containing GABAA receptors.•SGE-516 has pharmacokinetic properties suitable for oral dosing.•SGE-516 has anticonvulsant activity in both in vitro and in vivo epilepsy models.
