•The protein expression profiles of razor clam during CFPS was detected by iTRAQ.•27 differentially expressed proteins were identified.•Function of differentially expressed proteins were analyzed by GO and KEGG.•RT-qPCR revealed that H2A and S6K 2 alpha were post-transcriptional regulated.
