Abstract
Parkinson's disease is a chronic degenerative brain disease involving decreased level of dopamine generation and degeneration of substantia nigra. Online biological databases such as PDGene 2012 & 2014, ParkDB and NCBI GEO are built for revealing the potential mechanism of Parkinson's disease.The genetic expression data of microarray from brain tissue of patients with Parkinson's disease were collected. The construction of genetic network was made to analyze the protein-protein interaction by protein complex databases and molecular chemistry reaction database. The novel genetic influence and potential pathway may reveal the disease mechanism and cast the new concept of treatment strategy. The biological network analysis including centrality, clique analysis were used to analyze the interaction of mediator genes.The 29 candidate gene of PD were noted. Total QQPPI of genetic network contains 15 PPIs by POINeT. The possible mechanism of PD may involving high expression of RNA polymerase genes. Protein complexes such as RSmad and BRD4 complex are associated with PD. A clique including A2M, LRP1 and APOE is associated with dementia and plays important role in PD. SNCA may compromise the MAPK1-related pathways, and it may play a key role of the decrease of dopamine level in PD.The systematic biological analysis of biomarker and molecular network help to reveal the complexity of chronic brain disease and cast new era of potential pathways and treatment strategy of Parkinson's disease.
