•Extrapyramidal disturbances (ED) are side effects consequent to haloperidol treatment.•ED occurs due to dopamine receptor blockade in the nigrostriatal system.•ED are related to cholinergic activation and glutamatergic excitotoxicity.•Mg2++ supplementation inhibits Ca2++ and blockade NMDA receptors.•Mg2++ supplementation protects against haloperidol-induced movement disturbances.
