•K. lactis glycerol 3-phosphate dehydrogenase is required for growth on ethanol.•KlICL1 and KlMLS1 expressions are down-regulated in the Klgpd1 deletion.•This regulation is mediated by the transcription factors KlSip4 and KlCat8.•Both their promoter binding and activation functions are modulated.•KlGpd1 acts independent of the SNF1 complex in sensing the cells redox state.
