•The replication of PEDV strain 85-7 is extremely sensitive to IFN-β treatment.•HSP27 is an intracellular anti-PEDV factor depending on the IFN-β signaling pathway.•The ISGs Mx1 and Mx2, but not Viperin, show efficient inhibition of PEDV replication.•The down-regulation of HSP27 mediates PEDV escaping from host antiviral mechanism.
