•DBP can lead to the reduction of testosterone production and the inhibition of Cx43.•Cx43 is not essential for testosterone production in spite of its prominence in Leydig cells.•The change of testosterone production can regulate Cx43 expression.•DBP does not influence testosterone secretion via its effect on Cx43.
