•A sensitive, specific, and reproducible PRRSV RT-qPCR assay was established.•First report the replication kinetics of LP- and HP-PRRSV strain in MARC-145 cells and PAMs.•HP-PRRSV (Hu-N4) replicated to a higher titer than LP-PRRSV (CH-1a) in vitro.
