•MNHEPs could be stained with SYGO more clearly than before; Wright-Giemsa stain.•MN-incidences by DEN were comparable between this method and collagenase-using one.•Integrating the liver MN assay into toxicity studies can become easier by our method.•The liver MN assay can be done even after the completion of animal examinations.
