•We generated recombinant vectors using the Mycobacterium bovis BCG vaccine strain.•We vaccinated primates with recombinant BCG vectors expressing SIV Gag.•Primates had baseline humoral and cellular immune responses to mycobacteria.•Vaccinated primates generated strong CD8 T cell responses to SIV Gag.•Responses to SIV Gag were independent from baseline mycobacterial immunity.
