•We evaluated the intracellular phosphoprotein in response to growth factors or cytokines.•There was no characteristic signaling abnormality diagnostic for MDS.•MDS cases were often associated with more signaling aberrancies in more cellular populations.•Higher CD34+CD117 response to FL and SCF was frequently associated with high risk features in MDS.
