•Drosophila melanogaster Hox transcription factor Sex Combs Reduced (Scr) is spontaneously deamidated at residue Asn 321.•NMR spectroscopy is used to detect and quantify deamidation.•Deamidation reduces the affinity of Scr for target DNA.
