•Monodisperse magnetic polymer microspheres were developed.•The microspheres were coated with poly(ethylene glycol).•Properties of the microspheres were characterized by a range of methods.•DNA amplification in the presence of microspheres was investigated by qPCR.•Optimal coating of the microspheres was found.
