A physical map of human chromosome 5

Chromosome 5 comprises approximately 5% of the human genome or 170 Mb. As part of the Human Genome Project, we are constructing a high resolution physical map and cloned DNA contig of this chromosome. Assembled reagents include: (A) a natural deletion panel enabling the division of the chromosome into 30 bins, (B) 180 radiation hybrids containing chromosome 5 fragments, and (C) a chromosome 5-specific, arrayed cosmid library. All markers of interest are initially assigned to a bin and then the assignment further refined using radiation hybrid (RH) mapping techniques. In addition to two-point analysis of the RH data, chromosomal fragment assignment for each RH hybrid and the hybridization of pooled IRS-PCR fragments to arrayed libraries will be used to increase the map resolution using the inner product mapping method. Contigs will be assembled based on the STS content and hybridization results. This ongoing mapping effort is to include all known genes on chromosome 5, a large number of polymorphic loci and approximately 1200 STSs. The current radiation hybrid map includes all CEPH-Genethon and CHLC polymorphic markers enabling the immediate alignment of the chromosome 5 physical and genetic maps.
