•An antibody against extracellular domain of human claudin-4 was established.•Claudin-4 expression was associated with cancer progression in bladder cancer.•The antibody abrogated tight junction to leak intratumoral growth factors.•The antibody enhanced permeability of cisplatin into tumor.•The antibody increased anti-tumor effect of cisplatin in animal models.
