14634 Background: Inhibition of angiogenesis is a promising approach to treat cancer. Endostatin, the COOH-terminal proteolytic fragment of the basement membrane component collagen XVIII, represents one of the most potent endogenous angiogenic inhibitors. Recombinant human endostatin adenoviruses (Ad-hE), encapsulated with (Ad-hE/Lipo) and without liposome, were evaluated for their potential anti-tumor activity. Methods: MTT colorimetric assay was used to evaluate the inhibition of endostatin on human umbilical vein endothelial cells (HUVECs). An in vitro model was established for the observation of angiogenesis using primary culture of CT26 tumor tissues. Mice bearing CT26 colon cancer either by subcutaneous or lung metastatsis, were treated with different concentrations of Ad-hE or Ad-hE/Lipo by intratumor or vena caudalis injection respectively.
