锰对不同月龄大鼠脑黑质纹状体线粒体DNA"4834"缺失的影响

Abstract
线粒体DNA(mitochondrial DNA,mtDNA)约16 kb,占细胞总DNA的0.1%~0.5%,负责编码线粒体氧化磷酸化中4种复合酶的13个亚基.人的mtDNA长度约为16 569 bp[1],大鼠的mtDNA约为16 300 bp[2].mtDNA4977缺失在mtDNA的突变中最为常见,又叫"常见缺失(common deletion,CD)",是一段在mtDNA 13 447~13 459和8 470~8 482[3]核苷酸位点的顺式重复序列.与人类常见的mtDNA4977缺失相对应,大鼠比较常见的mtDNA缺失是mtDNA4834缺失;它的缺失也严重影响了线粒体呼吸链酶复合体的功能.本实验应用PCR方法对不同月龄大鼠mtDNA4834缺失进行比较研究,同时观察锰的影响.
