The functionality of mitochondrial membranes depends on the balanced accumulation of proteins and phospholipids. The endoplasmic reticulum serves both as donor membrane of precursor lipids and acceptor membrane for phospholipids synthesized within mitochondria, pointing to intense intermembrane lipid transport. The identification of mitochondrial lipid transfer proteins and of protein complexes tethering mitochondrial and ER membranes now provides insights into the molecular mechanisms of these phospholipid trafficking events.
