165-P: HLA-DRB1 AND -DQB1 ALLELE DISTRIBUTION BASED ON A SOUTH BRAZILIAN POPULATION OF BONE MARROW DONORS

Abstract
Aim The aim of this study was to determine the alelle frequency of HLA-DRB1 and -DQB1 in a bone marrow volunteers’ donors from South Brazilian population in order to contribute to the improvement of HLA alelles frequency knowledge in the Brazilian population. Methods HLA-DRB1 and –DQB1 allele frequency of 813 individuals recruited as bone marrow volunteers’ donors from South Brazilian population were analyzed.The sample ethnicity was determined by given information of individuals at the recruitment moment. Thus 82.2% were Caucasian, 1% Mestizos (mixed race), 0.9% Black, 0.4% Oriental, 0.2% Amerindian and 15.4% did not informed their ethnicity. Allele specific determination were done for loci HLA-DRB1 and -DQB1 using the commercial kit PCR-SSP Allele Specific (One Lambda Inc., Canoga Park, CA) following provider’s recommendation. Allele frequencies were calculated using the SPSS 17.0 statistical software. Results Fifty-six DRB1 alleles and eighteen DQB1 alleles were found. The most frequent alleles for DRB1 locus were DRB1 ∗ 07:01 (15.3%), DRB1 ∗ 03:01 (10.8%), DRB1 ∗ 15:01 (8.5%), DRB1 ∗ 13:01 (8.1%), DRB1 ∗ 01:01 (7.3%), DRB1 ∗ 11:01 (6.2%) and for DQB1 locus were DQB1 ∗ 03:01 (15.4%), DQB1 ∗ 02:02 (12.7%), DQB1 ∗ 05:01 (12%), DQB1 ∗ 02:01 (10.9%), DQB1 ∗ 06:02 (10.1%). Conclusions Because of the Caucasian predominance in our population sample, we could find a correlation between our findings and those early described in the literature for HLA-DRB1 and -DQB1 allele frequency for Caucasian population. These data can provide a better understanding of HLA-DRB1 and -DQB1 alleles frequencies which, in turn, will strengthen the ability of matching algorithms to predict allele matched donors for patients requiring a transplant.
