•Validation studies of the in vivo rat alkaline comet assay in liver and stomach.•Studies coordinated by Japanese Center for the Validation of Alternative Methods.•2-Acetylaminofluorene, azidothymidine, cisplatin, and isobutyraldehyde, tested.•AZT, AAF, and CPN were positive in at least one tissue.•IBA was negative in both tissues.
