Abstract
目的 探讨自体脂肪基质血管成分(SVF)阴茎海绵体注射对大鼠高血压性勃起功能障碍(ED)的影响及其可能机制.方法 选择30周龄健康雄性自发性高血压大鼠(SHR)40只,同系正常血压大鼠(WKY) 20只,采用无创血压仪测量大鼠尾部收缩压,颈部皮下注射阿扑吗啡(APO)检验大鼠阴茎勃起功能.将有ED的SHR大鼠分为2组:ED-SHR-SVF组(自体SVF阴茎海绵体注射治疗的高血压性ED大鼠,8只)和ED-SHR-磷酸盐缓冲液(PBS)组(PBS阴茎海绵体注射治疗的高血压性ED大鼠,8只).测定各组大鼠阴茎海绵体内压(ICP),采用Western印迹和RT-PCR法检测阴茎海绵体组织内皮型一氧化氮合酶(eNOS)蛋白和mRNA表达.结果 SHR大鼠尾部收缩压高于WKY大鼠[(197.47±6.82)比(125.23 ±4.65) mmHg,P<0.05].SHR大鼠阴茎勃起率为60%(24/40),WKY大鼠为100% (20/20).5V电刺激后ED-SHR-SVF组ICP明显高于ED-SHR-PBS组[(83.42±3.21)比(52.37 ±3.11) mmHg,P<0.05];ED-SHR-SVF组eNOS蛋白和mRNA表达明显高于ED-SHR-PBS组(0.43±0.03比0.18 ±0.05、0.92±0.05比0.41 ±0.06,均P<0.05).结论 高血压可导致大鼠ED,自体SVF可改善高血压性ED大鼠阴茎勃起功能,其机制可能与大鼠阴茎海绵体eNOS表达上调相关.
