Abstract
目的 研究CXC趋化因子受体3(CXCR3)单克隆抗体(mAb)对MCF-7人乳腺癌细胞和HepG2人肝癌细胞体外增殖及迁移的影响.方法 诱生腹水法制备CXCR3 mAb;流式细胞术筛选高表达CXCR3的MCF-7细胞和HepG2细胞;MTT法检测CXCR3mAb对MCF-7细胞和HepG2细胞增殖及干扰素诱导的T细胞α趋化因子(I-TAC)对MCF-7细胞和HepG2细胞促增殖的影响;TranswellTM法研究CXCR3 mAb对MCF-7细胞和HepG2细胞迁移及I-TAC对MCF-7细胞和HepG2细胞促迁移的影响.结果 MCF-7细胞和HepG2细胞CXCR3的阳性表达率分别为83.5%和96.2%;50 μg/mL CXCR3 mAb能够显著的抑制MCF-7细胞和HepG2细胞的增殖并抑制I-TAC对MCF-7细胞和HepG2细胞的促增殖作用;50 μg/mL CXCR3 mAb可抑制MCF-7细胞和HepG2细胞的迁移,并抑制I-TAC对MCF-7细胞和HepG2细胞促迁移作用.结论 CXCR3 mAb能够显著地抑制高表达CXCR3的肿瘤细胞的增殖及迁移.
