Abstract
目的:探讨黄连解毒汤乙酸乙酯提取物(ethyl acetate extract of Huanglian Jiedu decoction,EAHD)对白念珠菌菌丝形成的影响.方法:以0,78,312,1 250 mg·L-1 EAHD干预白念珠菌6h,采用倒置显微镜、荧光显微镜及扫描电镜分别观察白念珠菌菌丝形态;固体培养基上观测菌落形态;实时荧光定量PCR (qRT-PCR)检测白念珠菌菌丝形成相关基因HWP1,ALS3,UME6,SUN41,CSH1,CaPDE2的表达量.结果:312,1 250 mg·L-1EAHD可显著抑制白念珠菌菌丝的形成及菌落的形态;qRT-PCR检测显示1 250 mg·L-1EAHD使HWP1,ALS3,UME6,CSH1表达分别下调4.13,3.64,2.46,2.75倍,SUN41表达上调7.26倍,CaPDE2表达无变化.结论:EAHD可能通过抑制HWP1,ALS3,UME6,CSH1等相关基因的表达来抑制白念珠菌菌丝的形成.
