•A protease-insensitive region of SLR1 was identified by limited proteolysis.•An expression and purification system of the SLR1 GRAS domain has been established.•The purified SLR1 GRAS domain has a folded structure and binding activity to GID1.•A direct interaction between GID1 and GRAS domain has been detected.•A cysteine-less mutant SLR1 GRAS domain appears amenable to structural analyses.
