•Phylogenetic approach used for selection of key amino acids.•Mutations resulted in heat stability and improved allosteric properties of AGPase.•Combining several amino acid changes had an additive effect on heat stability.•Resulting enzyme (SH2-E) was heat stabile and no longer dependant on the activator.
