•PKR can be activated by glucolipitoxicity and pro-inflammatory cytokines in β-cells.•Activated PKR inhibited β-cell proliferation by arresting cell cycle at G1 phase.•Activated PKR fully abrogated the pro-proliferative effects of IGF-I on β-cells.
