Abstract
•O-linked mannosylation of glycoengineered Pichia pastoris-rhG-CSF was characterized.•We demonstrated that Thr134 is the only O-link glycosylation site using tandem MS.•The structure of O-mannosylation was determined by HPAEC-PAD and MS.
