•We identified a homologue of mammalian C1q in hagfish serum.•Hagfish C1q bound to various microbes in a Ca2+ dependent manner.•Hagfish C1q bound to peptidoglycan in a Ca2+ dependent manner.•Hagfish C1q bound to agarobiose but not to monosaccharide.•High density of agarobiose was necessary for stable binding of Hagfish C1q.
