•Isoform-specific antibody against murine type VI 3β-HSD (Hsd3b6) was created and characterized.•Adrenal Hsd3b6 is localized within the zona glomerulosa cells, where aldosterone is produced.•Testicular Hsd3b6 is localized within the interstitial Leydig cells, where testosterone is produced.•Dermal Hsd3b6 is localized within the cells composing the sebaceous glands.•Placental Hsd3b6 is localized within the giant trophoblast cells, where progesterone is produced.
