精子基因表达谱研究初探

Abstract
目的应用限制性显示技术,收集正常成年男性的精子基因表达谱探针,制备相应的精子表达谱芯片,以对成年男性正常精子的基因表达情况有一全面、系统的了解.方法提取成年男性正常精子的总RNA,逆转录为cDNA,经限制性内切酶酶切、加接头、PCR扩增、转化、克隆、鉴定等操作,收集成年男性正常精子的基因表达谱探针,制备相应的精子表达谱芯片,并进行初步的杂交分析.结果收集了1 859个基因片段,取其中368个基因探针制备芯片,进行了小量初试.结论证实在精子中有大量基因表达;提示我们自行制备的精子基因表达谱探针也具有较好的特异性和可信性.
