白血病骨髓基质增强HL-60细胞抵抗IDA化疗药物研究

Abstract
为了研究造血微环境异常在残留白血病发生中的作用和机制,采用Dexter型骨髓培养体系形成白血病骨髓基质细胞贴壁层,接种HL-60细胞共培养,用去甲氧基柔红霉素(IDA)处理,观察HL-60细胞的活性变化.结果表明:随着IDA剂量的增加及培养时间的延长,HL-60细胞活性逐渐减弱,与白血病骨髓基质共培养的HL-60细胞数明显高于正常对照组(P＜0.05),骨髓基质细胞层或单纯基质细胞条件培养液体外使IDA对HL-60细胞杀伤能力减弱.结论:白血病骨髓基质有助于HL-60细胞抵抗化疗药物,对残留白血病的发展具有一定作用.
