•DDB2 was subjected to SUMO modification in a UV-dependent manner.•PIASy was identified as a major SUMO E3 ligase for DDB2.•PIASy knockdown hampered the repair of CPDs, but not that of 6-4PPs.•PIASy interacted with DDB2.•PIASy colocalized with CPD and DDB2 in a UV-dependent manner.
