•Disruption of Kiaa0319l function results in large nodular periventricular heterotopias.•Neurons in these heterotopias are generated in mid- to late-gestation.•Neurons in these heterotopias are destined to upper cortical laminae.•Transfected neurons in the cerebral cortex are located in their expected laminae.•KIAA0319L is another candidate dyslexia susceptibility gene that modulates neuronal migration.
