•TLR3, IRF3, and IFN-β function as proinflammatory factors.•Increased expression of TLR3, IRF3, and IFN-β after stroke in tGCI rats was found.•Chloroquine pretreatment of stroke inhibits TLR3, IRF3, and IFN-β expression.•Inhibition of TLR3 signaling by Chloroquine pretreatment may have a beneficial effect.
