Abstract
目的研究内皮eNOS基因表达与兔实验性血管痉挛的关系.方法将16只兔随机分成内皮剥脱+普通饮食组(n=8)和内皮剥脱+胆固醇饮食组(n=8).血管造影观察球囊内皮剥脱前后以及普通饮食和高胆固醇饮食喂养8周后麦角新碱诱发血管痉挛情况.原位杂交和Northem bloting法定位和定量检测各组血管eNOS mRNA表达情况.结果血管造影显示,内皮剥脱+胆固醇饮食组可诱导出局限性血管痉挛.原位杂交显示eNOSmRNA定位在血管内皮细胞胞浆,痉挛血管节段内皮细胞内表达减少,Northernbloting法定量显示痉挛血管节段eNOSmRNA相对表达量较其他组明显减少.结论高胆固醇和内皮剥脱使eNOS基因转录和翻译下调,血管内皮eNOS mRNA表达降低可能与实验性血管痉挛的诱发高度相关.
