•Salicylaldehyde isonicotinoyl hydrazone (SIH) is an effective cytoprotective agent.•Boronyl salicylaldehyde isonicotinoyl hydrazone (BSIH) is activated to SIH.•LC–UV/MS methods were developed and validated for analysis of SIH and BSIH.•BSIH activation was studied in vitro using LC–UV.•LC–MS method was used in an in vitro stability experiment and pilot in vivo study.
