•We submitted cows to a sub dose of cloplostenol on Day 6 of estrous cycle.•Blood progestenone and luteal volume changes were recorded during 48h.•Blood progesterone dropped during the first 24h followed by a rebound.•Luteal volume decreased during the first 24h and then returned to initial values.
