Abstract
OBJECTIVE: To identifiy and characterize of mitochondrial DNA and cytochrome b gene in Martes zibellina L. heart and to establish the specific molecular technique for the purpose of protection, exploitation and utilization in LIXIN WAN. METHODS: The mtDNA were extracted from fresh Martes zibellina L. heart and heart powder respectively, and the restriction fragment length polymorphism of mitochondrial DNA was used to construct their physical maps of mtDNA (mtDNA-RFLP) with 6 restriction endonuclease, the cytochrome b (Cytb) gene segment was amplified by PCR. RESULTS: The complete mtDNA was separated from samples, and the Cytb gene with 310 bp segments was amplified by PCR. The restriction fragment length polymorphism of mitochondrial DNA gene with 6 restriction endonuclease were different between poultry and Martes zibellina L. heart. CONCLUSION: The results reveal that both methods are usful for seeking out the genetic markers among the Martes zibellina L. heart and different species hearts effectively.
