Abstract
胸腺五肽(TP5)能增进机体免疫力,已被用做抗感染、抗肿瘤和抗艾滋病的治疗药物.今以胸腺五肽(TP5)为研究对象,采用离子交换、纳滤和结晶相结合的新工艺分离制备TP5,有效地克服了传统工艺污染大、成本高的缺点.新工艺条件为:将TP5粗肽溶液以低流速通过NH-1树脂柱,采用NH4Cl与NaCl溶液洗涤杂质,选择氨水为洗脱剂,回收率为90.51%,TP5纯度为96.65%;采用SPT-100-150的纳滤膜对离子交换洗脱液进行浓缩脱盐,脱盐率为98.21%,TP5损失率为1.34%;建立乙醇/氯化钠协同作用的低温静态结晶工艺,TP5的一次循环结晶收率为48.35%,其纯度达到99.12%.
