Abstract
目的 研究RhoC基因沉默对肝癌细胞Bel7402凋亡和迁移的影响.方法 以RNAi沉默Bel7402细胞Rhoc基因的表达,FACS和RT-PCR检测细胞凋亡和凋亡相关基因水平,细胞划痕损伤和软琼脂克隆形成试验检测细胞迁移和生长特性.结果 RNAi组细胞凋亡率明显高于细胞对照组,RNAi组Bcl-2表达水平明显低于细胞对照组,只有RNAi组可检测到Bax基因的表达.两对照组细胞划痕损伤在48 h内愈合,而RNAi组则不能修复.RNAi组软琼脂克隆形成能力明显低于两对照组.结论 RhoC基因沉默能够促进肝癌细胞Bel7402凋亡并抑制细胞迁移和非锚定生长能力.
