The research of resveratrol resuming spermatogenesis damaged by 2.5-hexanedione

Abstract
Objective: To study the possible effect of resveratrol for improving dyszoospermia induced by 2.5-hexanedione(2.5-HD). Methods: 40 male SD rats were randomly divided into 5 groups. Group A was normally raised. Group B, C, D and E were exposed to 1% 2.5-HD for 5 weeks, and then Group C, D, E respectively received intragastric administration of different concentrations of resveratrol, namely 0.4%, 0.8%, 1.6%, at doses of 5ml/kg everyday for 9 weeks. Physical signs, body weight gain and testis weight were comparatively observed. Numbers and diameters of seminiferous tubules and expression of c-kit protein on spermatogenic cell membrane were analysed by HE stain and immunohistochemistry. Results: 2.5-HD resulted in weak body, lax skin, dim color pattern, tardy body weight gain, atrophy testis and so on. Histology showed emarcid seminiferous tubules, stagnant obsolete spermatogonia and depressive expression of c-kit protein. Through oral administration of resveratrol, the 2.5-HD-induced physical signs were improved, close to normal rats. The body weight increasement sped up. The recovery of testis weight was more obvious. Histologically, aliquots of the seminiferous epithelia began to differentiate and the spermatogenesis and expression of c-kit protein resumed, however, different from normal group. The diameters and numbers of seminiferous tubules were closer to the normal group along with increasement of resveratrol administrated. Conclusion: Resveratrol could shift 2.5-HD-induced dyszoospermia and resume the spermatogenesis.
